The chloroplast transformation technique is a powerful tool to produce useful proteins for medicine, agriculture and environment. However, some foreign proteins were not detectable or were accumulated to very low levels in transformed chloroplasts. This lack of accumulation has been explained by either “protein instability” or “mRNA instability”. We recently found that the translation initiation is an additional important factor for stable accumulation of foreign proteins in chloroplasts. The coordination between 5’-untranslated region and coding region is the additional factor for efficient translation initiation of foreign mRNAs. For a stable accumulation of foreign proteins in chloroplasts, it is important to design mRNAs which consider the translation regulation mechanisms. Here, we describe several factors influencing the translation of introduced mRNAs.
