Nucleotide sequence and temperature-dependent expression of XgroEL gene isolated from symbiotic bacteria of Amoeba proteus

A gene from symbiotic bacteria coding for a groE analog was cloned from a genomic DNA library of the symbiotic bacteria constructed in bacteriophage lambda ZAP and its nucleotide sequence was determined. The clone contained a DNA segment with an open reading frame of 288 nucleotides coding for a groES analog, and a second reading frame of 1653 nucleotides coding for a 67-kDa protein, a groEL analog. The gene had a 72% sequence similarity with the heat-shock operons of Legionella sp., E. coli and Coxiella burnetii. When the X-bacterial gene was transferred into E. coli, it was efficiently expressed, and its expression level was much higher at 37-degrees-C than at 24-degrees-C.
