Opposite mating type gametes of Chlamydomonas reinhardtii were induced by nitrogen deprivation and were mixed together. The gametes first discarded their cell-wall and fused to form a zygote, as true protoplasts in somatic fusion. Nitrogen-fixing Azotobacters were also mixed with algal cells before gamete fusion and the algal gametes fused in the presence of bacterial cells. After fusion of gametes and maturing of zygotes, the algal cells were maintained on nitrogen and carbohydrate free medium for over three years and were used for electronmicroscopic investigations. The bacterial cells were always found inside some of the algal cells and in the intercellular space of algal cells, suggesting the first step in establishing endosymbiosis between nitrogen-fixing Azotobacters and unicellular green alga, Chlamydomonas reinhardtii.
