A genomic expression library was prepared from DNA of symbiotic bacteria in the xD strain of Amoeba proteus, from which an 8-kb DNA fragment containing the gene coding for the symbiont-synthesized 29-kD protein was isolated and cloned. The inserts were subcloned in an expression vector and transferred to E. coli cells in which the gene was expressed at a high level, as tested by analyzing the bacterial total proteins by gel electrophoresis and immunoblotting. The transformed bacteria exported the fusion protein into the culture medium.
