Relationship between the frequency of AGN codons for Ser and the directional mutation pressure in metazoan mitochondrial protein genes was analyzed. The frequencies of T and G at the codon third position of four codon boxes, representing directional mutation pressure, show positive correlations with AGNs for Ser, whereas the frequencies of A and C at codon third position of four codon boxes show negative correlation, suggesting that the strand specific nucleotide composition bias (GT/AC bias) should be a candidate ruling the number of AGNs for Ser. We propose that the GT/AC bias is the major driving force of reassignment of AGRs in metazoan mitochondria.
